Overview of Gene Research
Dynein axonemal heavy chain 3 (DNAH3) resides in the inner dynein arm (IDA) of sperm flagella and is highly expressed in the testis. It is crucial for sperm motility as axonemal protein complexes like IDA drive sperm movement [1,2,3].
Multiple studies on humans and mice have shown the significance of DNAH3 in male fertility. In humans, biallelic variants of DNAH3 were identified in infertile men with asthenoteratozoospermia or oligoasthenoteratozoospermia. These variants led to deficient DNAH3 expression in sperm flagella, abnormal sperm flagellar morphology, and disrupted flagellar ultrastructure [1,2,3]. In Dnah3 knockout (KO) male mice, they were infertile, with severely reduced sperm movement, abnormal IDA and mitochondrion structure [1]. Non-functional DNAH3 expression decreased the expression of IDA-associated proteins in the spermatozoa flagella of both patients and KO mice [1].
In conclusion, DNAH3 is essential for sperm motility and male fertility. Studies using Dnah3 KO mouse models have clearly demonstrated its role in maintaining normal sperm flagellar structure and function, contributing to our understanding of male infertility caused by asthenoteratozoospermia or related sperm motility disorders [1,2,3].
