Overview of Gene Research
Asz1, also known as GASZ, is a crucial component of the piRNA pathway, which plays a vital role in repressing retrotransposons to safeguard genomic integrity in the male germline [1]. This gene is essential for germ cell and gonad development and is involved in regulating processes related to spermatogenesis and male fertility [2].
In Asz1 -null gonocytes of mice, MIWI2, responsible for introducing DNA methylation to retrotransposons in a piRNA -dependent manner, disappears from the nuclei of fetal gonocytes. This leads to upregulation of retrotransposons targeted by the piRNA pathway and non-annotated chimeric transcript variants in neonatal Asz1 -/- mice. These chimeras are formed by joining exons from retrotransposons and canonical genes. The retrotransposons inducing these aberrant transcripts are not fully methylated, consistent with the impaired nuclear localization of MIWI2 [1]. In zebrafish, asz1 mutant fish develop as sterile males with severely underdeveloped testes lacking germ cells. Germ cells in asz1 mutant juvenile gonads undergo extensive apoptosis, indicating Asz1 is essential for germ cell survival. Mechanistically, zygotic Asz1 is not required for primordial germ cell specification or migration but is essential during post-embryonic gonad development, likely by suppressing germline transposons [2].
In conclusion, Asz1 is essential for repressing retrotransposons and their associated aberrant chimeric transcripts in gonocytes through the piRNA pathway. Its absence leads to disrupted germ cell and gonad development, highlighting its significance in spermatogenesis and male fertility, as demonstrated by gene knockout models in mice and zebrafish [1,2].
