Overview of Gene Research
Parvb, also known as beta-parvin, is an actin-binding and focal adhesion protein. It is involved in linking integrins and associated proteins with intracellular pathways regulating actin cytoskeletal dynamics and cell survival [3]. The ILK-β-parvin pathway, in which Parvb is a part, can enhance cell motility [2].
In a PARVB cKO mouse model where the Parvb gene was deleted from proximal tubular epithelial cells, depletion of Parvb significantly attenuated cisplatin-induced renal tubular injury, including cell death and inflammation. Mechanistically, Parvb associates with TAK1, and its depletion promotes TAK1 degradation, inhibits TAK1 downstream signaling, and alleviates cisplatin-induced tubular cell damage [1]. In addition, in Fscn2 knockout mice, upregulation of Parvb contributed to cochlear cell death by inhibiting the ILK-AKT pathway [4].
In conclusion, Parvb plays a crucial role in regulating cell survival and motility through its involvement in various pathways. The study of Parvb using gene knockout mouse models has revealed its significance in diseases such as cisplatin-induced kidney damage and genetic hearing loss, providing potential therapeutic targets for these conditions.
