FNBP1, also known as Formin-binding protein 1, is a membrane-bound protein widely expressed in eukaryotic cells. It can stimulate F-actin polymerization beneath the curved plasma membrane, playing roles in cell migration, endocytosis, and is also involved in the non-canonical Wnt signaling pathway, which is crucial for vertebrate gastrulation [3]. It has been implicated in various biological processes and diseases, suggesting its overall biological importance. Genetic models, such as gene knockout (KO) or conditional knockout (CKO) mouse models, would be valuable for in-depth functional studies of FNBP1.
In cervical cancer cells, knockdown of FNBP1 leads to attenuation of FAK/PI3K/AKT signaling, causing significant apoptotic accumulation and proliferative inhibition, indicating its key role in maintaining the activity of focal adhesion kinase (FAK) and promoting cell survival [1]. In EMT-type gastric cancer cells, ablation of FNBP1 results in the loss of invasive ability, especially in a three-dimensional culture system, highlighting its importance in high-level cell motility [2]. In osteosarcoma cells, silencing LINC00987 inhibits cell proliferation, migration, and invasion by sponging miR-376a-5p to regulate FNBP1 expression [4].
In conclusion, FNBP1 is essential in multiple biological processes. Model-based research, especially through KO/CKO mouse models (if applicable in the studies), has revealed its significance in diseases like cervical cancer, gastric cancer, and osteosarcoma.
