Overview of Gene Research
Dmc1, short for disruption Meiotic cDNA 1, is a meiosis-specific recombinase. Meiotic recombination, which is crucial for fertility in most sexually-reproducing organisms, involves the repair of programmed DNA double-strand breaks. Dmc1, along with RAD51, is essential in this process as they search for and invade an intact DNA molecule on the homologous chromosome [1,2,3,5].
In Arabidopsis, Dmc1 attenuates RAD51-mediated recombination. Ectopic expression of Dmc1 in somatic cells makes plants hypersensitive to DNA damage and impairs RAD51-dependent homologous recombination. Also, in meiosis, a dominant-negative Dmc1 protein impairs RAD51-mediated DSB repair, suggesting Dmc1 acts to prevent RAD51-mediated recombination and this requires local assembly of Dmc1 nucleofilaments [1]. In mice, germ-line-specific conditional knockout (cKO) of FIGNL1 led to defective chromosome synapsis and impaired meiotic DSB repair with the accumulation of RAD51/DMC1 on meiotic chromosomes, indicating FIGNL1's role in homologous recombination at a post-assembly stage of RAD51/DMC1 filaments [4]. Similarly, both FIGNL1 and FIRRM are required for completing meiotic prophase in mouse spermatocytes, and the FIGNL1-FIRRM complex limits RAD51 and DMC1 accumulation on intact chromatin [7]. In wheat, CRISPR-Cas9-generated Dmc1-D1 mutants showed a significant decrease in crossovers at both high and low temperatures, indicating Dmc1-D1 is responsible for preserving normal crossover formation under such temperature conditions [6].
In conclusion, Dmc1 is vital for meiotic recombination, playing a key role in processes like preventing RAD51-mediated recombination, facilitating chromosome synapsis and crossover formation. The use of gene knockout and conditional knockout mouse models, as well as other genetic models like the wheat CRISPR mutants, has significantly enhanced our understanding of Dmc1's functions in meiosis and its potential implications in maintaining fertility, especially in the context of temperature-related effects on crops like wheat [1,4,6,7].
