Overview of Gene Research
Ppp1r14b, protein phosphatase 1 regulatory subunit 14B, is a poorly defined member of the protein phosphatase 1 regulatory subunits. It can inhibit protein phosphatase 1 (PP1) and various PP1 holoenzymes, which are crucial proteins in regulating cell growth, the cell cycle, and apoptosis. PPP1R14B is involved in multiple cellular pathways, and its abnormal expression is associated with various diseases, especially cancers [1,2].
In triple-negative breast cancer, PPP1R14B was upregulated, promoting cell proliferation, colony formation, migration, invasion, and paclitaxel resistance both in vitro and in vivo. Mechanistically, it maintained phosphorylation and stability of STMN1, a microtubule-destabilizing phosphoprotein [1].
In uterine corpus endometrial carcinoma, higher PPP1R14B expression was associated with poorer overall survival, disease-specific survival, and progression-free interval [2].
In prostate cancer, it was a potential diagnostic and prognostic biomarker, with its overexpression related to tumor immune inhibition, proliferation, and migration [3].
In hepatocellular carcinoma, PPP1R14B was highly expressed, promoting tumorigenesis by maintaining the stability and phosphorylation of RPS6KA1 and positively regulating the AKT/NF-κB pathway [4].
In cervical and endometrial cancers, PPP1R14B knockdown repressed cell proliferation and facilitated cell death by inhibiting the Akt signaling pathway [5].
In kidney renal clear cell carcinoma, upregulated PPP1R14B was an independent risk factor for cancer progression, and its knockdown reduced cell proliferation and migration in vitro. It was also associated with tumor-infiltrating immune cells [6].
In conclusion, PPP1R14B plays a significant role in cancer progression, often promoting tumor-related processes such as cell proliferation, migration, and drug resistance. Its involvement in multiple cancer types makes it a potential therapeutic target. Research on PPP1R14B using gene knockout or conditional knockout models, as inferred from the findings on its functional impacts, helps in understanding its role in cancer-related biological processes, thus contributing to the development of new cancer treatment strategies.
