Overview of Gene Research
Ldhd, lactate dehydrogenase D, is an enzyme that catalyzes the oxidation of D-lactate to pyruvate [3]. It plays a crucial role in the metabolism of D-2-hydroxyacids, and is associated with D-lactic acidosis [3]. Genetic models, such as gene knockout in mice, can be used to study its function.
In esophageal squamous cell carcinoma (ESCC), the CDK7-YAP-Ldhd axis promotes D-lactate elimination and ferroptosis defense, which supports cancer stem cell-like properties [1]. High Ldhd status is connected with poor prognosis in ESCC patients [1].
In hepatocellular carcinoma, Ldhd plays an important role in the prognosis of patients. Knockdown of Ldhd in HepG2 and Huh7 cells promoted cell proliferation, migration, and invasion, while overexpression showed the opposite effects [2].
In patients with neurological manifestations and mitochondrial complex IV deficiency, mutations in Ldhd lead to human D-lactate dehydrogenase deficiency and elevated serum D-lactate levels [4].
In conclusion, Ldhd is essential for D-lactate metabolism. Studies using knockout models have revealed its significant roles in various diseases, including ESCC and hepatocellular carcinoma, highlighting its potential as a therapeutic target for these diseases.
