Overview of Gene Research
Tent5a, also known as FAM46A, is a cytoplasmic poly(A) polymerase. It is involved in non-canonical polyadenylation processes, which play crucial roles in regulating gene expression at the post-transcriptional level. These processes are essential for various biological functions, including the production of secreted proteins and the regulation of mRNA stability [2,3].
In a Tent5a knockout (KO) mouse line, there are defects in bone formation. The KO mice display bone fragility and skeletal hypomineralization due to quantitative and qualitative collagen defects, as Tent5a is responsible for the cytoplasmic polyadenylation of mRNAs encoding Col1α1, Col1α2, and other secreted proteins involved in osteogenesis [2].
In another study, knockdown of Tent5a in C2C12 cells (a myoblast cell line) inhibited cell proliferation, migration, and the maturation of type I muscle fibers, both in vitro and in vivo. This was associated with decreased myogenin expression, suggesting Tent5a's role in muscle fiber formation in the context of adolescent idiopathic scoliosis [1].
In conclusion, Tent5a is essential for proper bone formation and muscle fiber maturation. The use of Tent5a KO mouse models has revealed its critical functions in these biological processes. These findings contribute to our understanding of the molecular mechanisms underlying bone-related diseases and adolescent idiopathic scoliosis.
