Overview of Gene Research
Nup210, also known as Nucleoporin 210 or gp210, is a key component of the nuclear pore complex (NPC) in eukaryotic cells. The NPC regulates nucleoplasmic transport, transcription, and genomic integrity. Nup210 is involved in various biological processes, and its study using genetic models like knockout mouse models provides insights into its functions [1,3,4,5].
Knocking down Nup210 in CRC cells inhibits cell proliferation, invasion, and metastasis in vivo and in vitro, and also reduces nuclear size, nuclear plasma material transport capacity, and NPC number and density on cell surface, indicating its role in promoting CRC progression [1]. In cervical cancer, knocking down Nup210 restores cell apoptosis and proliferation, and miR-22, a regulator of Nup210, is downregulated in cervical cancer, leading to Nup210 overexpression [2]. Nup210 knockout mice show delayed muscle repair after injury, abnormal muscle fiber distribution with age, and decreased muscle endurance, suggesting its importance in maintaining skeletal muscle integrity and function [3]. In breast cancer mouse models, Nup210 depletion suppresses lung metastasis, as Nup210 functions as a cellular mechanosensor and its knockout leads to defective mechanotransduction [4]. Nup210 knockout mice also have altered CD4/CD8 T cell ratios, indicating its T cell-intrinsic function in peripheral T cell homeostasis [5].
In conclusion, Nup210 plays essential roles in multiple biological processes such as nucleoplasmic transport, cell proliferation, apoptosis, muscle repair and T cell homeostasis. Gene knockout models of Nup210 have revealed its significance in diseases including colorectal cancer, cervical cancer, and breast cancer, providing a foundation for understanding disease mechanisms and potential therapeutic strategies.
