Overview of Gene Research
Mmgt1, also known as membrane magnesium transporter 1, is a key component in magnesium (Mg²⁺) transport. It belongs to the "membrane Mg²⁺ transporters" (MMgTs) family, with proteins residing in the membrane. MMgT1 mediates Mg²⁺ transport, playing a crucial role in maintaining Mg²⁺ homeostasis, which is essential for numerous cellular functions. Its expression in various cells suggests a wide-spread significance in different biological processes [2].
In Mycobacterium tuberculosis (Mtb) infection, Mtb-infected MMGT1-deficient macrophages promoted a switch to persistence, upregulated lipid metabolism genes, and accumulated lipid droplets during infection. Targeting triacylglycerol synthesis reduced both droplet formation and Mtb persistence. The orphan G protein-coupled receptor GPR156 is a key inducer of droplet accumulation in ΔMMGT1 cells, uncovering the role of MMGT1-GPR156-lipid droplets in the induction of Mtb persistence [1].
In conclusion, Mmgt1 is vital for Mg²⁺ transport and homeostasis. Its deficiency in macrophages reveals its role in regulating Mtb persistence through the MMGT1-GPR156-lipid droplet axis, providing insights into the mechanisms underlying Mtb infection and potentially new strategies for treating tuberculosis.
