Overview of Gene Research
Rhag, the gene encoding Rh-associated glycoprotein, is essential for the assembly of the Rh protein complex in the erythrocyte membrane and for the expression of Rh antigens [3]. It is also implicated in maintaining the structure and stability of erythrocytes [2]. Rhag may play a role in gas transport, with studies suggesting its major contribution to the NH₃ conductance of erythrocytes [1].
Mice with inactivated Rhag genes (Rhag-/ -) have a total absence of Rhag and Rh proteins [4]. These knockout mouse models help delineate the antigenic, biochemical and functional abnormalities of red cells, and the impact on hematological parameters and erythropoiesis [4]. In humans, single missense mutations in RhAG can induce spherocytosis and deficiencies in Rh complex formation [2]. A novel missense mutation was found to incompletely disrupt Rh antigen expression and selectively knock out RhD antigen expression [2]. Also, certain mutations in RhAG underlie the extremely rare Rhnull phenotype, characterized by the absence of all Rh antigens on erythrocytes [5].
In conclusion, Rhag is crucial for Rh antigen expression and erythrocyte stability. Research using gene-knockout mouse models has revealed its role in antigen expression, red cell function, and in relation to rare blood phenotypes like Rhnull. Understanding Rhag contributes to a better comprehension of erythrocyte-related biological processes and associated disorders.
