Overview of Gene Research
Crygc, encoding γC-crystallin protein, is crucial for maintaining lens transparency. Mutations in Crygc are associated with congenital cataract, a major cause of childhood blindness. Crystallin proteins contribute to the refractive properties of the lens, and their proper function is essential for normal vision [1-10].
Mouse models have provided valuable insights. For instance, mice with a dominant mutation in Crygc causing cataracts could be rescued by injecting Cas9 mRNA and a single-guide RNA targeting the mutant allele into zygotes. This demonstrated the potential of using the CRISPR-Cas9 system to correct genetic diseases related to Crygc [1]. Additionally, various mutations in Crygc, such as p.Trp131Arg, p.C130fs, p.R48H, p.E128*, etc., have been identified in different human families, expanding the understanding of cataract heterogeneity and the role of Crygc mutations in cataract formation [1, 3-5].
In conclusion, Crygc is essential for maintaining lens transparency, and its mutations are closely associated with congenital cataract. Mouse models, especially those using gene-editing techniques like CRISPR-Cas9, have been instrumental in understanding the role of Crygc in cataract-related diseases and exploring potential treatment strategies.
