Overview of Gene Research
Cfap298, also known as C21orf59, is a gene crucial for the transport/assembly of ciliary dyneins [2]. It is conserved in organisms with motile cilia and participates in the cytoplasmic preassembly of various ciliary dyneins, a process essential for cilia motility [2,3]. Motile cilia play critical roles in development and fertility, and thus Cfap298 is of great biological importance [2]. Genetic models, such as zebrafish mutants, are valuable for studying its functions.
In zebrafish, the cfap298tm304 mutant, with defective cilia motility and altered cerebrospinal fluid (CSF) flow, shows a 3D spinal deformity resembling adolescent idiopathic scoliosis (AIS) in humans [1]. This phenotype, more frequent in females, is evolutive during the juvenile phase, has a right convexity, a rotational component, and involves at least one dislocation [1]. Additionally, the reduction of cilia motility in cfap298tm304 mutants slows down CSF transport posteriorly in the central canal and abolishes the spontaneous activity of CSF-contacting neurons [4].
In conclusion, Cfap298 is essential for the cytoplasmic preassembly of ciliary dyneins and cilia motility. The study of cfap298tm304 zebrafish mutants has revealed its role in maintaining spine curvature and has provided insights into the development of AIS-like phenotypes, contributing to our understanding of the disease mechanism [1,4].
