Overview of Gene Research
Map2k6, also known as Mitogen-activated protein kinase kinase 6 (Mkk6), is a key component in the mitogen-activated protein kinase (MAPK) signaling pathway. It is involved in regulating various biological processes such as chromatin remodeling, cell proliferation, apoptosis, and stress responses. Dysregulation of Map2k6 has been linked to several diseases, highlighting its biological importance [1-10].

In somatic cell reprogramming, Map2k6 acts as a chromatin relaxer, enhancing the process. Its function in heterochromatin loosening and reprogramming requires kinase activity but is independent of its well-known target P38. It phosphorylates Gatad2b, which in turn elevates histone acetylation levels and loosens heterochromatin, promoting pluripotency gene expression [1].

In polycystic ovary syndrome (PCOS), circASPH promotes KGN cells proliferation through the miR-375/MAP2K6 axis, and circ_0005925 promotes granulosa cell growth by targeting miR-324-3p to upregulate MAP2K6, suggesting its role in PCOS-related cell growth regulation [2,6].

In ovarian cancer, MAP2K6-FP enhances the sensitiveness of paclitaxel via inducing autophagy [3].

In nasopharyngeal carcinoma, high MAP2K6 expression is associated with radiation resistance and poor prognosis [4].

In LPS-induced C28/I2 chondrocyte injury, circRNA-MSR regulates the process through the miR-643/MAP2K6 signaling pathway [5].

In hepatocellular carcinoma, APEX1 promotes oncogenicity via regulation of MAP2K6 [7].

In gut stem cell aging, mTORC1 increases MKK6 protein synthesis, activating the p38 MAPK-p53 pathway, leading to stem cell and villus aging [8].

In cell senescence, lncRNA MAGI2-AS3 affects the process via the ROS/MAP2K6/p38 signaling pathway [9].

In conclusion, Map2k6 plays essential roles in diverse biological processes and is closely associated with multiple diseases. Functional studies, especially those using in vivo models like KO/CKO mouse models (although not explicitly mentioned in the provided abstracts but commonly used in such research), could further clarify its detailed mechanisms in these biological processes and disease conditions, providing potential therapeutic targets for related diseases.
