As Cas9 can be programmed to make DNA double-strand breaks (DSBs) in the genome of cells, these breaks are then repaired by the cells’ DNA damage repair mechanisms, such as non-homologous end joining (NHEJ) or homologous recombination (HR). However, NHEJ-mediated repair of Cas9-generated breaks can generate small insertions and deletions at the targeted site during double-stranded break (DSB) repair. Cyagen’s CRISPR technology offers a solution to the limitations presented by the occurrence of NHEJ - our unique fertilization procedure increases the proportion of DSBs repaired by HR. This technique has greatly improved the accuracy and efficiency of CRISPR-based gene modifications and enabled more complicated models, including large fragment knockin (up to 15kb) and conditional knockouts.
