Overview of Gene Research
Slc6a4, encoding the serotonin transporter protein, is crucial for regulating serotonin (5-HT) levels, as the 5-HT transporter it codes for plays a key role in 5-HT reuptake [1,2,5]. Serotonin is involved in multiple physiological and psychological processes, and thus Slc6a4 is of great biological importance in relation to mental health and related functions [1,2,5].
Many studies focus on its polymorphisms and associations with various disorders. For example, the 5HTTLPR polymorphism in the 5' region of Slc6a4 has been widely studied in relation to psychiatric traits, with some suggesting associations with increased risk of posttraumatic stress disorder, higher likelihood for depression, obsessive-compulsive disorder, increased hostility and criminal behaviour [1,7]. Also, its DNA methylation has been associated with stress-related physiological changes; higher average levels of Slc6a4 DNAm were associated with higher average levels of salivary alpha-amylase (SAM axis) between-person, and with lower levels of salivary cortisol and alpha-amylase within-person [4]. In addition, in schizophrenia patients, the methylation level of Slc6a4 was significantly higher in women than in men, and higher in the schizophrenia group than in the control group, especially in the male subgroup [2]. However, meta-analysis did not find an association between Slc6a4 promoter and intron 2 variants and the risk of temporal lobe epilepsy, though the intron 2 VNTR seems to have opposite effects in different populations [3]. Also, no association was found between the Slc6a4 promoter polymorphism and alcohol dependence [6]. Moreover, the Slc6a4 short/short allele was found to increase anxiety symptoms in adolescents exposed to childhood food insecurity [8].
In conclusion, Slc6a4 is vital for regulating serotonin levels and is associated with numerous mental health-related conditions. Research on its polymorphisms, methylation, and gene-environment interactions has provided insights into the pathophysiology of various psychiatric and stress-related disorders, contributing to our understanding of the underlying mechanisms and potentially guiding future therapeutic strategies.
