Overview of Gene Research
Chka, or choline kinase α, is an enzyme that converts choline to phosphocholine. It is involved in the Kennedy pathways, which catalyze the de novo synthesis of phosphatidylcholine and phosphatidylethanolamine, key components of eukaryotic cell membranes [1].
Bi-allelic variants in CHKA can cause a neurodevelopmental disorder with epilepsy and microcephaly. These variants reduce the enzymatic activity of CHKA, impairing the first step of the Kennedy pathway [1]. In cancer research, CHKA is highly expressed in lung adenocarcinoma, colorectal carcinoma, and glioma, and its elevated expression is associated with poor prognosis. Knockdown of CHKA inhibits the proliferation, invasion, and migration of cancer cells [2,3,5]. In addition, CHKA SNP hCV1562388 genotypes with at least one C allele are associated with a reduced risk of spina bifida [4].
In conclusion, Chka is crucial for the Kennedy pathways in cell membrane synthesis. Studies, especially those on gene variants and knockdown in relevant disease models, have revealed its significance in neurodevelopmental disorders and various cancers, highlighting its potential as a biomarker and therapeutic target.
