Overview of Gene Research
Smad2, a member of the SMAD family, is a key intracellular mediator in the transforming growth factor-β (TGF-β) superfamily signaling pathway. TGF-β superfamily member signals are conveyed via cell-surface serine/threonine kinase receptors to Smad2. Once activated, Smad2 translocates from the cytoplasm to the nucleus to regulate gene expression, playing a crucial role in numerous biological processes such as cell growth, differentiation, and apoptosis [2].
In the context of diseases, studies have shown various roles of Smad2. Polystyrene microplastics can cause liver fibrotic injury in mice by activating the ROS/TGF-β/Smad2/3 signaling axis, leading to macrophage extracellular traps formation [1]. In melanoma, the NEAT1/miR-200b-3p/Smad2 axis promotes tumor progression by activating the EMT signaling pathway and immune responses [3]. In rheumatoid arthritis, over-expression of Smad2 inhibits pyroptosis of fibroblast-like synoviocytes and secretion of inflammatory factors via the TGF-β pathway [4]. Also, coffee's methylxanthine caffeine might slow fibrogenesis in liver diseases by inducing Smad2 degradation and impairing TGF-β signaling [5].
In conclusion, Smad2 is a vital regulator in the TGF-β signaling pathway, significantly influencing multiple biological processes. Research on Smad2, especially through in vivo models like mouse studies, has provided insights into its role in diseases such as liver fibrosis, melanoma, and rheumatoid arthritis. Understanding Smad2 function can potentially offer new therapeutic targets for these diseases.
