Overview of Gene Research
Gasdermin D (Gsdmd) is a key executor in pyroptosis, a form of programmed cell death. Inflammatory caspases, such as caspase-1,-4,-5 and-11, cleave Gsdmd, which is essential for pyroptosis induction. This process is involved in innate defenses, and Gsdmd-related pyroptosis has significance in host defense, inflammatory and autoimmune diseases [1,3].
Genetic KO of Gsdmd in mice alleviated depression-like behaviors and inhibited pyroptosis-associated protein expression, indicating its role in astrocyte pathological injury in depression [2]. In obstructive nephropathy, Gsdmd deletion in neutrophils protected the kidney from fibrosis as Gsdmd-dependent neutrophil extracellular traps promote macrophage-to-myofibroblast transition and renal fibrosis [4].
In conclusion, Gsdmd is crucial in pyroptosis, playing significant roles in diseases like depression and renal fibrosis. The use of Gsdmd KO mouse models has been instrumental in revealing its functions in these specific disease conditions, providing insights into disease mechanisms and potential therapeutic targets.
