Overview of Gene Research
Ano6, also known as TMEM16F, is a member of the TMEM16 protein (anoctamin) family. It has functions in ion transport, phospholipid scrambling, and regulation of other membrane proteins. Specifically, it is a major contributor to phosphatidylserine translocation from the inner to the outer leaflet of the plasma membrane and is associated with anion/cation channels activated by high Ca2+ concentrations [3]. It is involved in various biological processes and has been linked to multiple diseases.

In the context of disease, in gastrointestinal stromal tumors (GISTs), ANO6-plasmid inhibits proliferation, induces apoptosis, pyroptosis, and ferroptosis, and inhibits GIST growth in vivo, suggesting it could be a therapeutic target [1]. In breast cancer, lower ANO6 expression is seen in cancer tissues, but its overexpression predicts poor overall survival, and it may affect cancer progression via stroma-related pathways and macrophage polarization [2]. In glioma, ANO6 promotes cell proliferation and invasion through regulating the ERK signaling pathway [4]. In pancreatic ductal adenocarcinoma (PDAC), CAF-expressed ANO6 is essential for CAF trogocytosis, promoting PDAC cell survival and immune suppression, and its blockade may be a new PDAC therapy strategy [5]. In Alzheimer's cells, ANO6 targets TMEM30A to regulate endoplasmic reticulum stress-induced lipid peroxidation and ferroptosis, reducing neuronal loss injury [6]. A pan-cancer analysis also indicates ANO6 may serve as a biomarker for prognosis and melanoma progression [7].

Overall, Ano6 plays diverse and crucial roles in multiple biological processes and disease conditions. Studies using various models, though not specifically KO/CKO mouse models in these references, have revealed its significance in cancer development, neurodegenerative diseases, and other pathological states, providing potential therapeutic targets.
