Ppt2, or palmitoyl protein thioesterase 2, is a lysosomal thioesterase. It is highly associated with metabolism [1,2].
In fungi like Candida albicans, Ppt2 (phosphopantetheinyl transferase) transfers the phosphopantetheinyl group of coenzyme A to the acyl carrier protein Acp1 in mitochondria for lipoic acid synthesis essential for fungal respiration, making it a potential antifungal drug target [3,5].
In mouse models, disruption of Ppt2 causes an unusual lysosomal storage disorder with neurovisceral features. All Ppt2-deficient mice showed a neurodegenerative phenotype with spasticity and ataxia by 15 months, along with bone marrow infiltration, splenomegaly, and orange-brown pancreas due to lipofuscin pigment storage [4]. Ppt2-knockout mice developed spasticity at a median age of 29 weeks, and autofluorescent storage material was present in their brains [6].
In conclusion, Ppt2 is crucial for normal physiological function, especially in metabolism and lysosomal processes. Mouse knockout models have revealed its role in preventing a lysosomal storage disorder with neurovisceral manifestations. Understanding Ppt2 may provide insights into treating related neurodegenerative and fungal diseases.
