Overview of Gene Research
Cant1, also known as calcium-activated nucleotidase 1, is a phosphatase and a nucleotidase that preferentially hydrolyzes UDP to UMP and phosphate [3,4]. It is involved in glycosaminoglycan (GAG) synthesis and chondrocyte differentiation, and may play roles in multiple biological pathways such as cell cycle, DNA replication [1,2]. Gene knockout mouse models have been valuable in studying its functions [3].

In Cant1-knockout (KO) mice, which mimic Desbuquois dysplasia type 1, there are skeletal defects including short stature, thoracic kyphosis, and delta phalanx. The GAG content and extracellular matrix space are reduced in growth plate cartilage, and chondrocyte differentiation is impaired [3]. In addition, the synthesis and secretion of proteoglycans with shorter GAG chains occur in mutant mice, affecting the properties of decorin and aggrecan [4].

In conclusion, Cant1 is crucial for GAG synthesis and chondrocyte differentiation in cartilage. The Cant1 KO mouse model has provided insights into the pathogenesis of Desbuquois dysplasia type 1, highlighting the gene's importance in skeletal development and related diseases [3,4].
