Strain Description
Lysozyme (LYZ) gene encoded lysozyme is an antibacterial agent found in human milk, and also exists in the spleen, lung, kidney, white blood cells, plasma, saliva, and tears. The protein has antibacterial activity against a variety of bacteria. Mutations in LYZ are associated with hereditary renal amyloidosis. Lyz2 is the ortholog of LYZ in mice, and its expressed tissues include the hematopoietic system, liver, mesothelium, small intestine, and yolk sac.
Lyz2-IRES-iCre mice carry an insertion of the IRES-iCre expression element, and when this strain of mice is crossed with mice containing the loxP locus, sequence recombination between loxP loci mediated by Cre recombinase can occur in the bone marrow lineage cells (monocytes, mature macrophages, and granulocytes) of the offspring mice.
The homozygote Lyz2-IRES-iCre mice developed normally and were fertile.
The IRES-iCre element was inserted into the 3'UTR of Lyz2 in C57BL/6J mice.
Detection of Cre expression
a. Method
Lyz2-IRES-iCre mice were mated with Rosa26-LSL-tdTomato mice to produce double heterozygous offspring mice. Peripheral blood (PB), spleen, and bone marrow (BM) of the offspring mice were collected and fluorescence occurrence was observed by flow cytometry to confirm the expression of Cre protein.
b. Result
Experimental group (upper): Lyz2-IRES-iCre[KI/+];Rosa26-LSL-tdTomato[CKI/+]
Control group (bottom): Rosa26-LSL-tdTomato[CKI/+]
Description: Strong tdTomato fluorescence signals were detected in Cd11b+ cells in peripheral blood (PB), bone marrow (BM), and spleen.
c. Conclusion
This strain presents strong Cre-mediated recombination in Cd11b+ cells.
The Cre element of this strain is located on chromosome 10, please avoid using flox mice with the transgenic locus on chromosome 10 for breeding.
