Overview of Gene Research
SETBP1, also known as SET-binding protein 1/SEB/MRD29, is a SET-binding protein that regulates self-renewal potential through HOXA-protein activation [2]. It has significance in both neurological and cancer-related biological processes [1].
In myeloid neoplasms, mouse models have been crucial. For example, a mouse model expressing mutated SETBP1 in hematopoietic tissue showed profound alterations in the differentiation program of hematopoietic progenitors and developed a myeloid neoplasm with megakaryocytic dysplasia, splenomegaly, and bone marrow fibrosis [4]. Also, a Vav1-iCre;Setbp1fl/fl murine model indicated that SETBP1 depletion in normal hematopoiesis minimally alters self-renewal, differentiation, or reconstitution in vivo, and SETBP1 is dispensable for the development or maintenance of AML [3].
In conclusion, SETBP1 is important in regulating self-renewal potential via the HOXA-protein activation pathway. Mouse models, such as those with SETBP1 mutations or depletion, have revealed its role in myeloid neoplasms, showing its impact on hematopoietic progenitor differentiation and myeloid neoplasm development, as well as its dispensability in normal and malignant hematopoiesis.
