Overview of Gene Research
Zcchc10, zinc finger CCHC-type containing 10, is emerging as a crucial gene with significant implications in various biological processes and disease conditions. It is involved in regulating important pathways, such as those related to tumor suppression, potentially through its interaction with key proteins like p53 [1,2]. Genetic models can be highly valuable in further elucidating its functions.
In lung adenocarcinoma, Zcchc10 expression is lower in cancer tissues compared to adjacent non-cancerous tissues. Ectopic expression of Zcchc10 in lung cancer cells with wild-type p53 suppresses cell proliferation, colony formation, migration, invasion, and cisplatin resistance in vitro, as well as tumor growth and metastasis in vivo. Knockdown of Zcchc10 in normal lung cells has opposite effects. Zcchc10 binds and stabilizes p53 by disrupting the interaction between p53 and MDM2, thus exerting tumor-suppressive effects [1].
In acute myeloid leukemia (AML), Zcchc10 expression is downregulated. The lncRNA SNHG1 promotes ZCCHC10 promoter hypermethylation, leading to its silencing. Overexpression of Zcchc10 in AML increases p53 expression, suppresses cell proliferation and survival, and enhances sensitivity to venetoclax in a xenograft mouse model [2].
In colorectal cancer, miR-410-3p targets Zcchc10, and knockdown of Zcchc10 activates the NF-κB pathway, promoting epithelial-mesenchymal transition (EMT), cell migration, and invasion [3].
In conclusion, Zcchc10 is an important gene involved in tumor-suppressive functions, mainly through stabilizing p53 and influencing related signaling pathways. The study of Zcchc10 using gene knockout or other loss-of-function models has provided valuable insights into its role in lung cancer, AML, and colorectal cancer, which may help in developing new prognostic markers and therapeutic targets for these malignancies.
