Tyw3 is a gene encoding a S - adenosylmethionine (SAM)-dependent methyltransferase. It is responsible for the formation of a tRNA modification known as wybutosine and its derivatives, which are crucial for accurate decoding in protein synthesis [2]. It is also associated with pathways related to insulin resistance and may be involved in the pathogenesis of diseases such as amyotrophic lateral sclerosis (ALS) [1].
In a genome - wide association study in the Chinese Han population, TYW3/CRYZ was identified as a susceptibility gene for ALS, suggesting that insulin resistance might be implicated in ALS pathogenesis [1]. A genome - wide association study on circulating resistin levels found that genetic variants in the TYW3/CRYZ locus may be involved in the regulation of circulating resistin levels, and the resistin - rising allele of a SNP in this locus was associated with an increased coronary heart disease risk [3]. In esophageal cancer, TYW3 was one of the five prognostic RNA - binding proteins identified, with its down - regulation in tumor samples [4].
