Overview of Gene Research
Pxn, also known as paxillin, is a key component of the mechanotransducing machinery. It is involved in focal adhesion assembly, integrin signaling, and cell-matrix connection. Pxn is related to various cellular functions such as cell contraction, migration, and is also associated with autophagy-related pathways [2,3,4].
In a DDX17-induced DDX17HKO mouse model, knockout of DDX17, which is related to PXN-AS1 alternative splicing, led to a significant reduction in lung metastasis. This indicates that the DDX17-PXN-AS1 axis may play a role in HCC metastasis [1]. In ovarian cancer, high expression of PXN promoted cell proliferation, migration, and invasion, potentially by suppressing the p110β/Vps34/Beclin1 pathway [4]. In breast cancer cells, ULK1/2-mediated phosphorylation of PXN at S32 and S119 weakened its homotypic interactions and liquid-liquid phase separation, impairing focal adhesion assembly and cell migration [2].
In conclusion, Pxn plays essential roles in cell-matrix interactions, mechanotransduction, and autophagy-related pathways. Mouse models, such as the DDX17HKO mouse model, have helped to reveal its significance in cancer metastasis. In ovarian and breast cancers, Pxn is involved in promoting or suppressing cell migration and proliferation, highlighting its importance in understanding cancer progression mechanisms.
