Overview of Gene Research
Ppp1cb, also known as the beta-type catalytic subunit of serine/threonine-protein phosphatase 1, is involved in multiple biological processes. It participates in the RAS/MAPK pathway, and is associated with the development of certain developmental disorders. Additionally, it plays a role in adipogenesis, hepatic de novo lipogenesis, and may be related to the regulation of glycolysis in lung adenocarcinoma [1,2,4,5,6].

Mutations in Ppp1cb have been identified in patients with Noonan syndrome-like disorders. For instance, a patient with Noonan syndrome had a known de novo PPP1CB variant, and an individual with a p.Pro49Arg mutation in PPP1CB presented with features similar to Noonan syndrome-like disorder with loose anagen hair and craniosynostosis [1,3]. In 3T3-L1 preadipocytes, depletion of PPP1CB suppressed differentiation into mature adipocytes, suggesting its role as an adipogenic activator [5,6]. In the context of insulin resistance in mice, WDR6 interacts with PPP1CB to enhance fatty acid synthases transcription, promoting hepatic de novo lipogenesis [2].

In conclusion, Ppp1cb is essential in various biological processes such as development, adipogenesis, and lipid metabolism. Its involvement in diseases like Noonan syndrome-related disorders and potential implications in metabolic and cancer-related conditions, as revealed through patient-based and cell-based studies, highlight its significance. Understanding Ppp1cb may offer insights into the mechanisms of these diseases and potential therapeutic strategies.
