Overview of Gene Research
Serpind1, also known as heparin cofactor II, belongs to the serine protease inhibitor family. Some serpins, including Serpind1, utilize glycosaminoglycans (GAGs) to enhance the inhibition of target proteases [2].
In epithelial ovarian cancer, Serpind1 expression is significantly elevated, and high expression is associated with poor prognosis. Serpind1 promotes the proliferation, migration, invasion, G1-to-S phase transition, and epithelial-mesenchymal transition of ovarian cancer cells, while inhibiting apoptosis by promoting phosphorylation in the PI3K/AKT pathway. Inhibition of Serpind1 expression in ovarian cancer cells has opposite effects, and adding the PI3K/AKT pathway inhibitor to Serpind1-overexpressing cells can reverse its promoting effect on the malignant biological behavior of ovarian cancer cells [1].
In glaucoma, Serpind1 shows significant upregulation and plays a vital role in glaucoma occurrences, being associated with eye transparency decrease and glucose metabolism [3].
In patients with systemic sclerosis-associated pulmonary hypertension, the expression of Serpind1 is increased in the survivor group compared to the non-survivor group [4].
In intrahepatic cholestasis of pregnancy, the expression level of Serpind1 is remarkably lower in ICP patients than in healthy pregnant females, and it has a negative correlation with total bile acid (TBA) [5]. Polycyclic aromatic hydrocarbons (PAHs) accelerate the development of arteriosclerosis by promoting the expression of miR-155 to downregulate Serpind1 [6].
In conclusion, Serpind1 is a serine protease inhibitor that can interact with GAGs. Through studies in various disease models, it has been shown to play important roles in cancer (such as epithelial ovarian cancer), glaucoma, systemic sclerosis-associated pulmonary hypertension, intrahepatic cholestasis of pregnancy, and atherosclerosis. These findings from different disease-related research help to understand its biological functions and the potential for it to be a biomarker or therapeutic target in these diseases.
