Strain Description
The IL4 gene encodes interleukin-4 (IL-4), a multifunctional cytokine primarily secreted by Th2 cells, mast cells, and eosinophils. IL-4 promotes Th2 cell differentiation, B cell class switching to IgE, and alternative activation of macrophages (M2 polarization), playing critical roles in allergic responses, anti-parasitic immunity, and tissue repair. IL-4 and its receptor IL-4R are key regulators of immune responses and inflammation; dysregulation of this pathway is associated with various pathologies, including allergic diseases (e.g., asthma, atopic dermatitis, allergic rhinitis), parasitic infections, and certain cancers [1-5].
The Il4 KO mouse, a gene knockout model generated by deleting exons 1–4 of the murine Il4 gene (homologous to human IL4), serves as a valuable tool for investigating the pathogenesis of allergic disorders (e.g., asthma, atopic dermatitis, allergic rhinitis), anti-parasitic immunity, and specific cancers, as well as for developing related therapeutic strategies.

Strain Strategy
The mouse Il4 gene consists of 4 exons, with the ATG start codon in exon 1 and the TAG stop codon in exon 4. This strain was generated by deleting the region spanning exons 1–4 via genome editing.
