Strain Description

The Olig1 gene, which stands for Oligodendrocyte Transcription Factor 1, encodes a basic helix-loop-helix (bHLH) transcription factor protein that functions primarily in the central nervous system (CNS), particularly in oligodendrocyte lineage cells (including oligodendrocyte precursor cells) and some neurons and radial glia. Its main function involves the differentiation and maturation of oligodendrocytes and the subsequent process of myelin formation and remyelination following injury in the adult CNS. Olig1 expression is crucial for the complete maturation of oligodendrocytes, with some studies suggesting a more significant role in the brain compared to the spinal cord, and its protein can be observed in the nucleus and cytoplasm depending on the cell stage and context (e.g., translocating back to the nucleus during remyelination).
Olig1-P2A-iCre mice were generated using gene editing technology to replace the TGA stop codon of the mouse endogenous Olig1 gene with P2A-iCre cassette. Under the control of the Olig1 gene's regulatory elements, the iCre recombinase (a codon-optimized Cre recombinase) is expressed. Using the 2A peptide for linkage avoids the drawbacks of low protein activity and low downstream gene expression in multi-gene expression processes. When Olig1-P2A-iCre mice are crossed with mice containing loxP sites, Cre recombinase-mediated recombination between loxP sites is expected to occur in the Olig1-positive cells in the offspring.
