Strain Description

IL-18 is an important pro-inflammatory cytokine that binds to its receptor complex (IL-18Rα encoded by IL18R1 and IL-18Rβ encoded by IL18RAP), activating downstream MyD88/NF-κB and MAPK signaling pathways and playing key regulatory roles in innate and adaptive immunity [1]. Abnormal activation of the human IL-18/IL-18R signaling pathway is closely associated with various inflammatory diseases, including inflammatory bowel disease (IBD), rheumatoid arthritis, psoriasis, metabolic inflammation, and regulation of the tumor immune microenvironment [2].
The huIL18R1/huIL18RAP humanized mouse model was generated using gene editing technology, in which the mouse Il18r1 gene region from exon 1 to the downstream region of exon 13 of Il18rap was replaced with the corresponding human IL18R1 gene region from exon 1 to the downstream region of exon 10 of IL18RAP, enabling the expression of the human IL-18Rα/IL-18Rβ receptor complex. This model can be used to mimic the functional roles of human IL-18/IL-18R signaling in inflammatory responses. Its applications mainly include mechanistic studies of inflammatory bowel disease (IBD), rheumatoid arthritis, psoriasis and skin inflammation, as well as metabolic disease-associated inflammation. In addition, this model can be used for exploring cancer immunotherapy strategies and evaluating the potential effects of various therapeutic approaches targeting the IL-18/IL-18R pathway in regulating intestinal inflammation, joint inflammation, skin inflammation, metabolic inflammation, and the tumor microenvironment.

Strain Strategy

The sequence from exon 1 of the mouse Il18r1 gene to the downstream of exon 13 of the Il18rap gene was replaced with the sequence from exon 1 of the human IL18R1 gene to the downstream of exon 10 of the IL18RAP gene. Since the replaced human genomic fragment contains the SDR42E1P5 pseudogene and MIR4772 microRNA gene, these human genomic elements were also introduced into the mouse genome as part of the humanized region.

Application Area

Inflammatory Bowel Disease (IBD) Mechanism Research: Used to investigate the pathogenic mechanisms by which human IL-18R signaling drives intestinal inflammation, epithelial barrier disruption, and Th1/Th17 immune responses in Crohn’s disease and ulcerative colitis;
Rheumatoid Arthritis Therapeutic Evaluation: Used to assess the inhibitory effects of neutralizing antibodies targeting human IL-18 or IL-18R, as well as recombinant IL-18 binding protein (IL-18BP), on joint inflammation and bone erosion;
Psoriasis and Skin Inflammation Research: Used to study the role of human IL-18R signaling in promoting keratinocyte activation and neutrophil infiltration in psoriasis-like dermatitis;
Inflammatory Research in Metabolic Diseases: Used to explore the mechanisms by which the human IL-18/IL-18R pathway drives macrophage activation and insulin resistance in obesity, type 2 diabetes (T2D), and metabolic dysfunction-associated steatohepatitis (MASH);
Tumor Immunotherapy Validation: Used to evaluate the therapeutic potential of IL-18R agonists or antagonists in modulating the tumor microenvironment and enhancing NK cell and T cell anti-tumor activity.
