Strain Description
Kallikrein-related peptidase 2 (KLK2, human kallikrein 2, hK2) and kallikrein-related peptidase 3 (KLK3, prostate-specific antigen, PSA) are members of the kallikrein-related peptidase family. Both are androgen-regulated serine proteases that are highly expressed in prostate epithelial cells. KLK3 encodes prostate-specific antigen (PSA), which is an important biomarker for prostate cancer (PCa) diagnosis and disease monitoring. KLK2 is involved in prostate cancer cell proliferation, invasion, and tumor microenvironment regulation. Altered expression or regulation of KLK2/KLK3 is associated with prostate cancer progression and therapeutic responses [1-3]. The mouse genome lacks direct orthologs of KLK2 and KLK3.
The H11-hKLK2/hKLK3 humanized mouse model was generated by inserting the Human KLK3 promoter-Kozak-Human KLK3 CDS-WPRE-BGH pA-2*beta-globin insulator-Human KLK2 promoter-Kozak-Human KLK2 CDS-WPRE-BGH pA cassette into the mouse H11 locus using gene editing technology. This model can be used to investigate the roles of human KLK2/KLK3 in prostate cancer development and tumor immunoregulation. Its applications mainly include the screening, development, and evaluation of KLK2/KLK3-targeted therapeutics and immunotherapy strategies, as well as prostate cancer disease mechanism studies. It can be used to evaluate therapeutic approaches targeting KLK2-related antigens, including CAR-T cell therapy, bispecific antibodies, and KLK3-associated cancer vaccines, for their potential roles in cancer treatment.
