Strain Description

Gpr84 (G protein-coupled receptor 84) encodes a Gi/o-coupled G protein-coupled receptor (GPCR) that senses metabolic signals such as medium-chain fatty acids and is believed to be involved in various physiological processes including nutrient sensing, immune regulation, and tissue homeostasis. Gpr84 is mainly expressed in myeloid cells such as neutrophils, monocytes, and macrophages. Its expression can be induced by inflammatory stimuli such as lipopolysaccharide (LPS), and it is involved in immune cell chemotaxis, phagocytosis, and the production of inflammatory cytokines [1]. Furthermore, GPR84 also has functions in tissues such as the gastrointestinal tract, adipose tissue, liver, and central nervous system, potentially participating in processes including intestinal hormone and neuropeptide release, energy metabolism, inflammation, and fibrosis [1-2]. Studies have shown that activation of GPR84 can regulate related signaling pathways such as Akt, ERK, and NF-κB, and enhance the inflammatory response and phagocytic activity of macrophages [3].
Gpr84-P2A-CreERT2-T2A-EGFP mice were generated by gene editing, in which the stop codon of the mouse Gpr84 gene was replaced with the P2A-CreERT2-T2A-EGFP gene expression cassette. In the absence of tamoxifen treatment, the CreERT2 recombinase mainly resides in the cytoplasm; upon tamoxifen treatment, CreERT2 recombinase translocates to the nucleus and exerts its recombination function. When Gpr84-P2A-CreERT2-T2A-EGFP mice are crossed with mice containing loxP sites, the offspring are expected to undergo Cre recombinase-mediated recombination between loxP sites in Gpr84-positive cells following tamoxifen induction.
