Overview of Gene Research
HYOU1, also known as Orp150, is an endoplasmic reticulum molecular chaperone protein. Various stimuli can trigger an unfolded protein response to endoplasmic reticulum stress, accompanied by the up-regulation of HYOU1 expression [2]. HYOU1 is involved in multiple biological processes and associated with many diseases.
In the context of intestinal injury, Fut2-dependent fucosylation of HYOU1 at the N-glycosylation site of asparagine (Asn) 862 was found to protect intestinal stem cells (ISCs) against inflammatory injury. Fut2ΔISC (ISC-specific Fut2 knockout) mouse models demonstrated that FUT2 depletion in ISCs aggravated epithelial damage and disrupted ISC growth and proliferation by escalating LPS-induced endoplasmic reticulum (ER) stress and initiating the IRE1/TRAF2/ASK1/JNK branch of the unfolded protein response (UPR) [1].
In conclusion, HYOU1 plays a crucial role in protecting cells from stress-related damage, as seen in the protection of ISCs against inflammatory injury. The use of gene-knockout mouse models, like the Fut2ΔISC mice, has been instrumental in uncovering these functions, providing insights into potential therapeutic strategies for intestinal injury disorders.
