Overview of Gene Research
Cyba, encoding p22(phox), is the gene for the cytochrome b558 alpha polypeptide. The p22(phox) protein is a crucial component of the superoxide-generating nicotinamide adenine dinucleotide phosphate (NADPH) oxidases (NOXs). It partners with NOX2 to form cytochrome b558 mainly in phagocytes, which is responsible for killing microorganisms during infections. Also, it combines with NOX1, NOX3 and NOX4, and the p22(phox)-NOX complexes are significant sources of reactive oxygen species (ROS) in tissues and cells, involved in various physiological and pathological processes [1].

The p22(phox)-deficient mouse strain nmf333 has been used to study the role of p22(phox) in inner ear balance in association with NOX3. However, the relevance of p22(phox) for NOX3 function remains uncertain as AR22(0)CGD patients do not have vestibular dysfunction [1]. In addition, studies on human genetic variations, like the C242T polymorphism of CYBA, have been conducted in relation to coronary artery and heart diseases, though results are conflicting [1]. CYBA genotypes associated with reduced ROS formation, such as rs1049254/G and rs4673/A, are related to unassisted ventilation, shorter recovery time in Guillain-Barré syndrome, reduced multiple sclerosis severity and delayed onset of secondary progressive MS [2,5]. The A allele of rs7195830 in CYBA is a risk allele for renal cell carcinoma, and high CYBA expression is associated with advanced tumour characteristics and worse prognosis [3]. The T allele of the CYBA C242T polymorphism is related to a decreased risk of preeclampsia, while its combination with certain SOD2 genotypes increases the risk [4].

In conclusion, Cyba is essential for the function of NADPH oxidases, playing a key role in immune defense against infections. Through model-based research, including mouse models, its role in various diseases such as cardiovascular, neurological, and renal cell carcinoma has been revealed. The genetic variations of Cyba are associated with different disease severities and risks, highlighting its importance in understanding disease mechanisms and potential biomarker discovery.
