F8a, a DNA sequence, has significance in multiple biological contexts.
In the context of hemophilia A, a copy of the F8A gene is located within intron 22 of the factor VIII (FVIII) coagulation gene, with two others 500 kb upstream. Homologous recombination between the intragenic F8A gene copy and either the distal or proximal copy leads to severe hemophilia A as no intact FVIII is produced [1].
In immunohistochemical studies comparing clear cell sarcoma of the kidney (CCSK) and Wilms' tumor (WT), CCSK cells were negative for Factor-8 related antigen (F8A), while neither blastemal, stromal nor epithelial elements in WT were positive for F8A. This can potentially aid in differentiating between these two tumors with different prognoses and treatment requirements [2].
In conclusion, F8a is associated with the molecular pathogenesis of hemophilia A through its role in the FVIII gene inversion mechanism. In tumor-related studies, the absence of F8A expression in specific cell types of CCSK and WT can be a useful diagnostic feature.
