Overview of Gene Research
Ccn2, also known as connective tissue growth factor (CTGF), is a matricellular protein. It plays diverse roles in biological processes, regulating cell growth, invasion, and migration. Ccn2 is involved in multiple pathways such as TGF-β, TGFBR/SMAD, ILK and Syk signaling cascades, and is crucial for the development of tissues like cartilage and bone in the skeletal system [1,2,4,5].
A monoallelic variant in CCN2 causes autosomal dominant spondyloepimetaphyseal dysplasia with low bone mass. Osteoblast lineage-specific Ccn2-deficient mice (Ccn2fl/fl;Prx1Cre) recapitulated phenotypes including low bone mass, with pathological mechanisms involving decreased bone formation, increased bone resorption, and abnormal growth plate formation, highlighting the critical role of CCN2 in osteogenesis [3].
In conclusion, Ccn2 is essential for tissue development, especially in the skeletal system. The study of Ccn2-deficient mouse models has revealed its role in skeletal dysplasia, providing insights into the mechanisms of bone-related diseases and potential therapeutic targets.
