Overview of Gene Research
Grik1, which encodes the GluK1 subunit of the excitatory kainate receptor, is essential for brain function and is involved in many mental and neurological diseases [3,4,5,6,7]. The kainate receptor is part of the ionotropic glutamate receptor family, and its signaling pathways are crucial for synaptic plasticity, learning, and memory processes [3,4].
In glioblastoma, knockdown of Grik1 retarded GBM cells growth, migration, and invasion, indicating that Grik1 promotes glioblastoma malignancy and may be a biomarker for diagnosis and therapy [2]. In endometriosis, while the study focused on lncRNA GRIK1-AS1, it's worth noting that GRIK1-AS1 deficiency accelerates endometriosis progression through epigenetic regulation mechanisms related to SFRP1 methylation [1]. In Attention deficit hyperactivity disorder (ADHD), Grik1 risk variants and Grik1 deficiency were detected in Indian ADHD probands, suggesting its role in ADHD etiology [4].
In conclusion, Grik1 is crucial for normal brain function and is implicated in multiple disease conditions such as glioblastoma, endometriosis, and ADHD. Functional studies, especially those involving loss-of-function experiments like knockdown in glioblastoma, have revealed its role in promoting disease-related cell behaviors.
