Overview of Gene Research
Il17a, a major member of the IL-17 cytokine family, is mainly produced by T helper 17 (Th17) cells, with other cell types like CD8+ T cells also capable of its production. It has a host-protective capacity in healthy individuals but excessive elevation is linked to autoimmune diseases and cancer, playing a role in immune-related pathways [3].
In a COPD mouse model, selective knockout of Il17a in the airway epithelium significantly reduced neutrophilic infiltration, peribronchial inflammation, pro-inflammatory mediators, and mucus production. This shows that Il17a, expressed in the airway epithelium, can act in an autocrine manner to amplify inflammation and increase mucus production in COPD pathogenesis [1].
In HLA-B27 transgenic rats, a peptide-based vaccine targeting Il17a alleviated the spondyloarthritis phenotype, inhibiting enthesitis, bone destruction, and new bone formation [2].
In conclusion, Il17a is crucial in regulating immune-related processes. The gene knockout mouse models in COPD and spondyloarthritis research have revealed its role in promoting inflammation and disease-related phenotypes, highlighting its potential as a therapeutic target in these diseases.
