Overview of Gene Research
Notch1 is a gene encoding a single-pass transmembrane receptor. The Notch signaling pathway, in which Notch1 is a key component, is crucial for cell-fate determination during development, regulating processes like tissue homeostasis, differentiation, and proliferation [3,5].
In hematological malignancies, NOTCH1 activating mutations are identified in over 50% of T-cell acute lymphoblastic leukemias and 12% of chronic lymphocytic leukemias, highlighting its role in leukemia pathogenesis [2]. In ischemic heart disease, aberrant Notch1 signaling is universal, affecting endothelial cells, vascular smooth muscle cells, and macrophages, as well as various remodeling stages [1]. In gastric cancer, the Notch1/CDH5 axis promotes cancer cell growth and metastasis [4].
In conclusion, Notch1 plays essential roles in development and disease. Its study, especially through gene knockout or conditional knockout mouse models (implied by functional studies in the references), has revealed its significance in diseases such as leukemia, ischemic heart disease, and gastric cancer, providing potential therapeutic targets.
