Overview of Gene Research
OR6A2, the human ortholog of mouse Olfr2, is an olfactory receptor belonging to the G protein–coupled chemoreceptor family. It plays a role in detecting the compound octanal, which is involved in the activation of the NLRP3 inflammasome and induction of interleukin-1β secretion in macrophages. This activation is associated with the inflammatory response, and is relevant to the development of atherosclerosis [1,2,3].
Genetic targeting of Olfr2 in mice (a form of gene knockout model) significantly reduced atherosclerotic plaques. Boosting octanal levels exacerbated atherosclerosis, indicating that the activation of Olfr2 (and by extension OR6A2) through octanal binding drives the progression of atherosclerosis [1].
In conclusion, OR6A2 is involved in the NLRP3-dependent IL-1 production pathway in macrophages. The gene knockout models of its mouse ortholog Olfr2 have revealed its crucial role in the development of atherosclerosis, suggesting that inhibiting OR6A2 may be a promising strategy for preventing and treating this disease [1].
