Pde8a, a high-affinity cAMP-specific phosphodiesterase, plays a crucial role in regulating cyclic adenosine 3',5'-monophosphate (cAMP) levels. cAMP is a key second-messenger involved in multiple signaling pathways, and Pde8a's function in cAMP degradation is essential for controlling these pathways. It is expressed in various tissues such as the ovary, testis, heart, nervous system, and osteocytes, suggesting its broad biological importance [2,3,4].
In the heart, AKAP12 upregulation associates with PDE8 to reduce total intracellular cAMP levels, accelerating cardiac dysfunction. Adult primary cardiomyocytes from AKAP12-overexpressing mice had reduced contractility and impaired calcium handling in response to isoproterenol, which was reversed by a selective PDE8 inhibitor. This indicates that Pde8a is involved in cardiac function regulation [1]. In Leydig cells from mice with a targeted mutation in the Pde8a gene, they are sensitized to the action of LH in terms of testosterone production, although in human studies, PDE8A was excluded as a significant PCOS candidate gene [2]. In osteocytes, mechanical-sensitive focal adhesion kinase (FAK) can modulate intracellular cAMP levels via PDE8A. Inhibition of FAK or PDE8A led to increased cAMP, PKA activity, and expression of cAMP-regulated target genes [4].
In conclusion, Pde8a is essential for cAMP-related signaling in multiple biological processes. Studies using gene-modified mouse models, like those with Pde8a targeted mutations, have revealed its role in cardiac function, androgen production, and osteocyte mechanosensing.
