Overview of Gene Research
POU2AF1, also known as OCA-B, is a transcription co-factor. It plays crucial roles in various biological processes. It is involved in immune-related regulation, such as in the differentiation of Tfh cells where it promotes Tfh formation and migration into germinal centers by upregulating CXCR5 [2]. It also functions in the regulation of host defense genes in the human airway epithelium, helping maintain a host defense tone even in pathogen-free conditions [4].
In overweight patients, POU2AF1 expression was upregulated in retroperitoneal adipose tissue and increased during adipogenesis. Overexpression of POU2AF1 promoted MSCs adipogenesis, while its silencing inhibited adipogenesis, suggesting its role in adipose-related biological processes through the HDAC1 signaling pathway [1]. In B-cell malignancies dependent on POU2AF1, treatment with mSWI/SNF ATPase degraders led to chromatin eviction of POU2AF1 and decreased downstream signaling, indicating its importance in these malignancies [3].
In conclusion, POU2AF1 is a key transcription co-factor involved in diverse biological processes, including immune regulation, adipogenesis, and host defense. Its study, especially through functional studies in relevant disease models, provides insights into its role in diseases such as adiposity-related disorders and B-cell malignancies, which may offer potential therapeutic targets.
