Overview of Gene Research
Aldh4a1, a member of the aldehyde dehydrogenase superfamily, is a key mitochondrial enzyme in the proline metabolism pathway. It catalyzes the final steps of both proline and hydroxyproline catabolism, converting L-glutamate-γ-semialdehyde to L-glutamate in proline metabolism and 4-hydroxy-L-glutamate-γ-semialdehyde to 4-erythro-hydroxy-L-glutamate in hydroxyproline metabolism [5,6,7].
A knockout mouse model of Aldh4a1 was constructed using CRISPR-Cas9 technology. Results showed that Aldh4a1^(-/-) male mice had normal spermatogenesis but defects in sperm maturation in the epididymis, with impaired motility, increased morphological abnormalities, and increased spontaneous acrosome reaction. Transmission electron microscopy revealed vacuoles in sperm mitochondria, fractures in sperm necks, and vacuoles, indicating that ALDH4A1 is vital for sperm flagellum structure and sperm maturation in mice [2].
In conclusion, Aldh4a1 plays a crucial role in proline and hydroxyproline catabolism and is essential for sperm maturation in mice. The Aldh4a1 knockout mouse model has provided valuable insights into its function in male reproduction. Additionally, ALDH4A1 has been implicated in other biological processes and diseases, such as atherosclerosis, schizophrenia, and muscle health, suggesting its broad-ranging importance in biology and medicine [1,3,4].
