Overview of Gene Research
Sry, the sex-determining region Y gene, is a master regulator of male sex determination in mammals. It is located on the Y chromosome and triggers the differentiation of the bipotential genital ridge into testes. Sry initiates a cascade of gene networks by directly regulating Sox9 expression, which is crucial for supporting cell differentiation, Leydig cell specification, vasculature formation, and testis cord development [2].
In mouse models, Sry expression is tightly regulated both spatially and temporally. Insufficient or delayed Sry expression can lead to XY gonadal sex reversal, highlighting its critical role in male development. Mouse Sry is the most intensively studied mammalian model for sex determination. Several transcription factors act as trans-acting factors for Sry expression, and epigenetic modifications of Sry also play a role as cis-acting factors in sex determination [1].
In conclusion, Sry is essential for male sex determination in mammals. Mouse models, especially those with gene knockout (KO) or conditional knockout (CKO) of Sry, have been crucial in revealing its role in sex determination and the consequences of its dysregulation, such as disorders of sex development in humans [1,2].
