Overview of Gene Research
Tlr4, a member of the Toll-like receptor (TLR) family, is a transmembrane receptor that plays a central role in the innate immune response. It recognizes lipopolysaccharide (LPS) or bacterial endotoxins, mediating inflammatory responses. Through adaptor proteins like MyD88 and TRIF, it activates transcription factors NF-κB and AP-1, leading to upregulation of pro-inflammatory cytokines and increased interferon expression [1,2].
In a study with TLR4-knockout (TLR4-/-) mice challenged with LPS, deletion of TLR4 attenuated LPS-induced acute liver injury by inhibiting inflammation and apoptosis via the TLR4/MyD88/NF-κB signaling pathway, demonstrating that TLR4 plays a vital role in septic injury [3].
In conclusion, Tlr4 is crucial for the innate immune response and inflammation regulation. TLR4-KO mouse models have shown its significance in septic acute liver injury, highlighting its potential as a therapeutic target for related diseases.
