Overview of Gene Research
Otulinl, also known as FAM105A, is the only putative pseudodeubiquitinase of the ovarian tumor protease (OTU domain) family in humans. It lacks deubiquitinase activity but may function through mediating protein-protein interactions. It localizes to the endoplasmic reticulum (ER) and nuclear envelope, and its interactome shows enrichment in proteins from ER/outer nuclear, Golgi and vesicular membranes [1].
The crystal structure of FAM105A (Otulinl) revealed structural deficiencies in the active site and substrate-binding infrastructure, which impairs normal deubiquitinase (DUB) function. It has no catalytic function against all ubiquitin linkages and cannot bind ubiquitin compared with catalytically active FAM105B/OTULIN [1].
In conclusion, Otulinl is a pseudodeubiquitinase that localizes to the ER membrane and likely functions through protein-protein interactions rather than deubiquitinase activity. Although the reference does not mention KO/CKO mouse models, understanding its non-catalytic functions can potentially contribute to insights in areas related to cell fate control and ubiquitin-mediated cellular processes [1,2].
