Selenoo, also known as selenoprotein O and SELO, is an evolutionarily conserved selenoprotein. It catalyzes a posttranslational protein modification called AMPylation, which is essential for the oxidative stress response [2]. SELENOO localizes to the mitochondria and AMPylates proteins involved in redox homeostasis [2].
In a murine model of spontaneous melanoma metastasis, SELENOO deficiency significantly reduced metastasis to distant visceral organs [1]. This effect could be rescued by treatment with the antioxidant N-acetylcysteine [1]. Mechanistically, SELENOO AMPylated succinate dehydrogenase subunit A, one of the key subunits of mitochondrial complex II, and SELENOO-deficient cells featured increased mitochondrial complex II activity [1].
