Overview of Gene Research
Klrk1, encoding NKG2D, is a homodimeric lectin-like receptor. NKG2D can bind to ligands expressed by cancer cells, playing a role in cancer immunosurveillance and antitumor effects. It is involved in the immune response pathway, and its abnormal expression may be related to the occurrence and development of diseases [1,2].
In lung adenocarcinoma, Klrk1 expression was lower in tumor tissues than in normal tissues. High Klrk1 expression was associated with improved overall survival and relapse-free survival, and in vitro studies indicated that it inhibited tumor cell proliferation and migration [1].
In head and neck squamous cell carcinoma (HNSCC), Klrk1 was highly expressed and correlated with better prognosis and immune cell infiltration, especially CD4/8(+) T lymphocytes. High expression levels of Klrk1 ligands, such as UL16 binding protein (ULBP) 1-3, were associated with an increased risk of death, and Klrk1 expression was negatively correlated with ULBP1-3 [2].
In the study of antithyroid drug-induced agranulocytosis, the KLRC4-Klrk1 gene polymorphism was associated with susceptibility and progression of the disease. Patients carrying the rs2734565-C allele had a higher susceptibility and faster onset time [3].
In conclusion, Klrk1 is involved in cancer-related immune responses and has potential as a prognostic biomarker in lung adenocarcinoma and HNSCC. The study of Klrk1 gene polymorphisms also helps understand the susceptibility and progression of certain diseases, providing insights into disease mechanisms and potential treatment strategies.
